今年踌躇满志地申请基金委的重点项目，希望可以支持“葡萄糖转运蛋白的结构与机理
”研究。我还一直志在必得，因为这个课题的重要性远大于我之前所有的研究，好像基
金委还从来没有立项支持营养物质跨膜转运这个很重要的基础研究领域，而且我们知道
是一定可以做出成果、对得起这份资助。

拍手叫好的有，妒忌讥讽者的也有。但事实上隔行如隔山，叫好者和讥讽者可能并不真
正了解对颜宁的工作。那么问题来了，颜宁的工作到底牛不牛？有多牛？颜值到底高不
高？
先要回答的问题是结构生物学到底有多牛？因为颜宁毕竟是做结构的。以最具说服力的
诺贝尔奖为例，百年化学+生理和医学诺奖215项中有13项是结构生物学相关的，占6%[1
]。尤其是其中1962年沃森克里克DNA双螺旋结构的发现，是近代生物学的基础，也是百
年来科学领域最重要的发现之一。再看美国霍华德休斯研究所（HHMI）资助的科学领域
。HHMI是全球最具声望的针对生物和医学的私立基金组织，他们资助的科学家个个含金
量十足。目前有超过300名科学家受到HHMI的资助，而其中40人从事结构生物学相关研
究[2]。抛开这些统计不说，结构是功能的基础，而生物大分子是所有结构中最具挑战

的太厉害，底部时间越长，就越危险，因为资金源源不断进来，mm就不停的上上下下乱
窜狂赚，所以他们自然希望的是底部震荡时间越久越好。说实话，看看09年，仔细看，
其实是去了3次30块。最后一次最猛烈，当时uco的价钱离前两个底又低了20%。飙哥如
果随便上网搜下当时的新闻包括一些分析，几乎跟现在一模一样，一直不停的说glut

Indeed, there are many Ph.D. programs that can be closed. The first one on
my list to close would be the Religion program in Columbia university. Why
do we need Ph.D.'s specializing in religion? Leave that area to the rabis
and ministers!

glut的callback直接处理硬件中断.
这是我个人的一点理解.
如果你看到WPARAM,peekmessage什么的,这用的是windows gui技术,会很慢.
典型的glut程序一般会有如下语句:
glutReshapeFunc(changeSize);//OK
glutKeyboardFunc(processNormalKeys);
理论上glut在windows或linux上的速度不应该有太大差异. |

前者是可逆交联，并且在组织内移动速度快。后者铰链度更大，并且不可逆，固定得更
好。但是移动速度更慢。而且对抗体保存不够好。现在一般采用混合固定，4%PFA加0.5
%的glut。
如果是大的组织，先初步固定之后，再slice成小块继续固定。
如果是免疫荧光，glut的浓度应当低于0.5% |

我对炼油股研究很少，不敢妄谈建议，不过个人感觉炼油股不能拿，因为现在成品油
glut应该比原油glut还要厉害，马上夏天的peak driving season也要结束了，汽油需
求会降低。
长线的话我推荐逢低吸纳大油股，BP，Shell,股息较高，被套了就当定存，压力不大，
油价总有一天会涨回去的，拿着1,2年也不是很怂。

的情况只要用PFA (4% paraformaldehyde)就差不多了。除非你对细胞结构要求很高，
要看胞内细胞器或者做电镜的，可以加一点glut。我以前试过加0.1% glut,再往上加抗
体就失效了

发帖数: 1089 | 35 固定组织需要考虑组织大小，否则不能彻底固定。如果组织小于1mm，2hr PFA足够了。
否则需要4度过夜固定。或者先固定2小时，再切成小块固定。
上面有人说0.1%的glut都会影响抗体结合。你可以自己试试，如果是这样的情况，可以
只用4% PFA。
固定完之后，需要dehydration和infiltration，再包埋切片。

2000年，颜宁在普林斯顿大学开始了博士生涯，师从如今的清华大学副校长施一公教授。2007年，年仅30岁的颜宁从普林斯顿大学学成而归，回到自己本科时的母校清华大学任教，成为当时清华最年轻的博士生导师。
“清华厚道、大气、稳重，普林斯顿优雅、淡定、高贵，这是我一生中最爱的两个地方。”颜宁曾经在接受媒体采访时这样表示。

5月8日，清华大学相关负责人向媒体证实了这一消息。该负责人表示，颜宁经本人慎重考虑并与学院和学校仔细沟通，已决定接受美国普林斯顿大学分子生物学系雪莉·蒂尔曼终身讲席教授的职位，将于近期前往就任该教职。
但与此同时，一篇名为《清华才女颜宁教授为何负气出走普林斯顿?》的文章在网上流传，作者雷歌撰文称，“看看颜宁在科学网的博客，再结合新华社简短的消息，就一目了然：颜宁是因为连续两年拿不到自然科学基金委的科研项目，负气而去。”
北京时间记者就此采访了清华大学相关部门，清华大学相关部门的工作人员告诉记者，颜宁本人目前不在国内，无法接受采访。并表示，颜宁已在微博就此事进行了回应。
北京时间记者看到，5月9日中午，颜宁在微博中回应称，否认自己去普林斯顿和基金会有关，相关文章“明明就是莫名其妙无稽之谈”，并表示自己在微信朋友圈第一时间辟谣了一篇类似文章。
