其实比较简单的办法就是 random_shuffle line[N]，N 是总共 data 的行数。
就算 N 是几百万，也是小意思。然后 sort 一下 line。然后就读一行，如果该
行不在 line[] 里，扔掉。在 line[] 里留下。这办法可以处理很大的文件。。。
上面是 sampling w/o replacement 。sampling w/ replacement 可以同理。
唯一麻烦的是，如果某 algorithm 对 order 比较 sensitive 。。。 |

之后我们实验室的很多人做transfection都不成功或者其他assay也有问题。然后老板
今天叫了某个公司的人来做mycoplasma test，测了我的2个细胞line和另外一个人的细
胞line。测出来的结果是所有的细胞line都被mycoplasma污染了，然后实验室陷入了极
大的恐慌。
大家纷纷在讨论过去这2个月细胞实验是不是白做了，然后还在讲我们department多少
人问我们借过细胞line，然后我们是不是把其他实验室的细胞也都污染了。还有一个
concern就是我前一阵子刚freeze了一些细胞stock，他们担心是不是所有放在一起的
cell stock也都污染了。
现在实验室还在恐慌之中，因为一切是从我的细胞总是死开始的，所以虽然大家没明说
，但是我觉得其实大家心里都觉得我的细胞是源头。我觉得我们实验室现在已经造成了
很大的损失，而且感觉都是我造成的，现在超级难过。

首先，现在认为从正常细胞变成cancer cell需要经过multiple steps。所以每一个不
同的stage都可以有细胞系。
1. 正常细胞。最常见的，比如我们的PBMC，fibroblast等，还有很多其他的细胞。当
然这样的正常细胞难以成为细胞系。因为会senescense，不能长期繁殖。不同species
的细胞能够繁殖的代数不同，人类的好像多数在10-20代左右（看donor的年龄等），但
是都不能无限繁殖。我们买到的，一般是经过几次传代扩展的low passage cell。不同
batch可能是不同的donor。
2. immortalized。正常细胞经过一些处理可以成为immortalized cell line。最简单
的，比如我们的PBMC用EBV感染一下，可以immortalize B cells。其他还有很多方法，
比如用chemical筛选等。immortalized cells不会senescense，可以无限繁殖，但是仍
保留部分正常细胞的性状。具体是那些，跟正常细胞区别有多大，要看具体的细胞系，

repeat）位点
大概三个月前测的是这个位点是homozygous有6个repeat
然后最近重新解冻另外一个stock，start cell culture，测量同样的位点，居然成了
heterozygous，一半仍然6个repeat，另外一半成了10个repeat；反复做了很多遍，换
了所有的reagent生怕是contamination，但结果还是一样。
问了lab technitian，这个neuroblastoma在我们实验室都是一样的来源，他也怀疑这
个repeat是不是dynamics的
三个月前的cell line的passage不记得了
最近这个cell line的passage大概是p10
我想问下，cancer cell line的genome microsatellite难道真的会传代传着传着就发

我在MEDICAL FLOOR作了一年半，刚去SURGICAL UNIT(GYNE-ONCOLOGY),病人普篇
INDEPENDENT，体力劳动没那磨重，但这个UNIT护士绝大部分是白人，有白人慢悠悠做
事的作分，有时稍范错误她们马上填上INCIDENT REPORT，这是我不喜欢的。另外，昨
天我范了一错误，我不知此错误是否大到足以令我辞职。
昨天一病人术后来我们UNIT，血压很低，我报告CHARGE NURSE， RESIDENT来了，医嘱
LACTATED RINGER 和NS，分别RUN BY GRAVITY， 病人有两个PERIPHERAL LINE，其中一
个LINE，我忘了PRIME （因SURGICAL UNIT 的TUBE与MEDICALFLOOR的不一样）我CLAMP
了此LINE BY ACCIDENT，NS不能流动，夜班护士发现报告了MANAGER。MANAGER找我谈话
了，说真话，我很尴尬的。
我知道是我错了，但人人都会范错误。我想问的是我这个错误（忘了PRIME PERIPHERAL

line（maximum line大概在coolant reservoir一半处，实际line在8成左右）。这会有
坏处吗，需不需要减少？
还有加engine oil, power steering fluid, automatic transmission fluid, brake
fluid等，因不知前车主加的是什么型号的油。如果混合加入不同型号的相应油会出问

接头很大上面似乎镀了chrome，插头镀金？dock一端还有两个卡子。
佛罗里达一个人卖的，似乎是他自己做的。
还是需要Line Out的，效果明显不一样。
耳放太菜了，回头重点升级一下。

我们的property line上占了0.9 feet的地, fence 的长度大概有342米, 延伸到后面的

直在翻修房子, 并且昨天刚好加了一个大门. 这样一来就相当于把我们的土地的占有完
全永久化了.我查了我们州的法律, 如果土地被nortoriously占七年, 不提出异议的话,
土地就归占有人所有了. 这样, 我就不希望今后我们在卖房子时遇到麻烦.
昨天, 我刚好见到了我邻居的男朋友, 是他一直在帮她做工程, 就将这件事情说了, 他
让我和他女朋友谈. 我的想法是:
(1)让她和我签一个协议, 明确我们还是遵守property line, 但是我不要他们移动, 拆
出现在的fence 和门. 但是一旦她卖房子或者是我们卖房子, 他们应该回到原来的

一般如果有 easement 你买房子的时候大概知道一些，你担心的是 gas line 的走向，
电线的走向，等，这些 easement 一般不能知道的。
这个 digsafe 就是帮你探测这些 lines 的，你如果做 permanent 的 shed, 最好做一
个。
但是我家装 shed 时就没做 digsafe，因为是后院，有 lines 的可能性太小，而且，
我的 shed 没有地基， 随时可以挪。 |

roof vent，现在又堵了，这下修不好了，snake只是通一个小缝，马上又会堵上，让我
们找jetter来clean管子里面的grease，查了一下价钱，至少400+。价钱不是主要
concern，问题是后院有一个sewer，我们打开井盖看到里面有脏水，记得两年前看过，
里面是干的，会不会是public sewer line堵了？所以我们家kitchen line才堵？请大
家帮忙解惑，我是应该叫county的人来看看，还是请sewer company的人来看看，还是

空调的condensation drain line里面老是听到滴水的噪音，主要是在空调刚开始工作
和工作结束之后的时候。
一开始我们还不太清楚，听上去的声音更像是有人在里面狠狠的敲墙。声音大到晚上已
经可以把人吵醒。找了专门修空调的人过来看，他们说drain line从主卧旁边的墙过，
听到的声音就是滴水声。但是我们觉得这个滴水声是在是吵的离谱，而且声音还不是很
统一，有的时候轻，有的时候很吵。
修空调的说要在furnace边上的墙上重新开一个口，放弃原来的drain line，直接从新
开的口出去，狮子大开口要1000刀。我想这就是把原来的PVC管子给切了，然后重新接
一个出去就要4位数？这个也太坑爹了。
另外觉得前几年也没有这种感觉，今年一入夏就觉得这个噪音特别明显，不知道版上的
各个大大能不能指点迷津？ |

收到了第一个月的账单(1个月+4天）
话费 83.99 费和税就付了 41.76. 税率49.7%
请问有家庭计划的， 你们也是这么高的税率吗？

他们说INQUIRY 是HARD HIT，不知道会损伤我的信用多少啊。我大概750左右啊，不要
太低啊，马上想买车呢。。。我还有2张信用卡，一张准备CANCEL（是AE的，马上要收
年费了），一张是不是也去要求加CREDIT LINE比较好，以前看书说是14天之内无论多
少个INQURIY都算一个HARD HIT，这样对CREDTI LINE的损伤不会太多？有这个说法吗？
另外一个想要求增加CREDIT LINE的是CHASE FREEDOM的，不知道他家给的大方吗？至少
给我MATCH一下BOA也行啊。。

credit line就会受限制 |

If this does not meet your needs, please discuss available credit line
options with our Customer Service Specialist when you call to confirm
receipt of your card.
大家会在收到卡给chase打电话确认的时候提出提高credit line吗？要提供什么理由比
较好呢？会再有hard pull吗？实际上现在给的credit line已经比预期的要高，也足够
用了，但既然有这个选项，还是想多多益善了。多谢各位！ |
