i run some eletrophoresis to see how the enzyme works on the plasmid DNA.
but something weird happen.
the control group (without digest enzyme) got 2 bands appear on the gel image
the digest group( all the 3 enzymes) got only 1 bands appear on the gel image
anyone has experience on this before?
anyone can give out any possible analysis on this?
i am just new to the lab, and my professor has no time to take care of me .

That's possible.
THe plasmid without digestion will generally have 3 bands based on the plasmid
configureation: the fastest one is supercoiled, the medium is linear ( both
strands of the plamsid were broken during extraction, and the slowest one is
the circular(one strand was broken). The quality determines the band pattern
in UNCUT plasmid.
You said only one band shows in 3-enzyme digestion. What is the size? You may
got four 2kb bands from a 8kb plasmid. BUT the most possible caus
