1. get a set of degenerate primers, usually designed specifically
for your organism.
2. design a specific primer which binds to one end of your DNA.
3. do 3 rounds of PCR, 3 minute elongation time, 10 minute extention time.
i) very low anealing temperature (35C?) for 25 cycles. take 1 ul,

都是小蛋白. 大点的蛋白有chaperon帮忙. 此外细胞内部是个crowding的环境.
Crowding对蛋白质folding有没有影响我不清楚. 不过纯粹的靠crowding就能fold DNA
到很小的尺寸. 这个还是很惊人的.

Why? some cell should die after a few passages of division. If they don't

invitrogen platium hifi. 3‘引物设计了6个，条件也优化（AT,elonging time, mg+
)，要不什么也没有要不没有P出目的条带（测序后错配了）。。。唉 都要崩溃了..
另外的一个基因到是P出来了，这个基因3'端有很多重复序 GC 50-60%的样子 请教大家
一般用什么办法或强大的酶。。。

温度已测好，57°C
用SYBR green做qPCR，SYBR新从BioRad订的
25μl total volume
12.5μl SYBR green master mix + 0.1μl primerF (0.1μM in the fianl
concentration)+ 0.1μl primerR + 50ng genomic DNA + H2O fill up to 25μl
用的PCR program如下：
95°C 10min + (95°C 30sec, 57°C 30sec, 72°C 30sec) 40cycle + dissociation
curve
72°C elongation step读数
结果没有任何信号，dissociation curve当然也没信号或者有一些非常弱且杂乱的信号
我把sample拿出来，跑了个2%agar gel，结果非常清晰干净的一条带，size完全正确，

还是不明白LZ的意思，如果mRNA的结构是cap-gene1-IRES-gene2-3UTR，gene1的翻译可
能被抑制，IRES一般含有stem loop高级结构，会阻碍translation elongation。如果
cap-gene1-3UTR, IRES-gene1-3UTR是两个独立的mRNA,gene1的表达不太可能被抑制，
因为IRES的翻译效率很低。
另外IRES不完全是3-tail independent。是否就完全5cap independent，我觉得也不好
讲。IRES只是说ribosom loading不需要cap（其实这个概念本身就问题多多），cap的
作用不限于ribosom entry，还包括mRNA 稳定等。 |

扩增全长基因，正是我需要检测到特定条件下的不同组织中的病毒基因的突变的差异，
按理讲我可以用DeepSeq，不过小实验室不太富裕，而且就三四个基因，本身也不太需
要，
我RT的elongation 时间一般是90分钟，
会不会是SuperIII的效率要比SuperII差些？
我本意是想问问各位，有没有什么trick可以更高效的RT扩增出特别稀少的模板，
