They concluded that Asc regulated Dock2 expression through controlling Dock2
mRNA stability.
They failed to provide Asc complementation experimental data which is
considered critical in a genetic approach.

入另一个strain。用的phage是transducing phage，据提供者说high frequency of
lysogenization。我用plaques提取lytic phage，然后和recipient混合，incubate 20
分钟以后立即稀释、涂板，结果没有一个带resistant marker的colony长出来。
我的实验方法可能会出什么问题？
这种phage是不是主要进行specialized transduction？收集lysogen然后transduce是
不是可行的方法？

work on PC. And a bad thing about them is that they were written by
programmers not biologists, so they are in general not intuitive, not
tailored to the needs of every biologist. Besides, usually you have to pay a

Kan slow down the growth a bit. That allows phage to catch up and clear out
your culture.
Can you use TonA knockout strain to do your experiments? if so, you can try
that. (You can get the strain from a number of vendors.) The reason is that
the popular bacteria phage contamination is due to T1 phage. The receptor of
it on E. coli is TonA.

这个到底推翻什么认识还是要细说。几条跟aging相连的通路都是比较成熟的（当然也
有细节上
的争执--但是生物不就是在于细节么）。而节食作为一个影响所有（所有!）激素和所
有细胞内通路的东西，其实一直有报道说某种节食在老鼠里不成功什么的。最近aging
cell报道说在一堆strain里面节食，一半寿命延长一半寿命缩短。
这个猴子主要是比较难养，活得太长了，我看见他们作者人数居然不是很多。大概
看了看figures好像没什么分子之类的，估计是这些data想留着再发几篇15分以上的
paper吧。
说实话我觉得猴子这种阶级分明的动物本来不就是大部分猴子处于经常性节食状态？节
食在老鼠里面做一般是定时定量给东西，那猴子这样岂不是大家都等猴王吃了才敢吃。
待会看看paper他们怎么解决这个问题。
SIRT1那个GSK确实是被人坑了呵呵～英国那个组negative results也是好不容易搞到
nature上了，当时他们搞这个课题，老做不出来，把一作和二作一个博士一个博后郁闷
得不行了。

129strain很容易的睾丸癌，肿瘤的发病率也比其它strains 高。而且性格极其温顺，
但胎生的每胎数量小（平均，5-8）。在野外它是无法生存的。但温顺的性格非常适合
实验室使用。其它特性不妨碍一般实验而已。 |

，比如LD50的量去晒，如果你用个LD99的量筛肯定得不到抗性strain啊。另外美国原则
上规定在GM crop的边上要有10－20%的refugee，就是常规非GM作物用来缓解害虫的环
境压力，从而减少和减慢抗性的产生。传统的杀虫剂要向控制refugee就太困难了。 |

一般来说从jackson lab向国内买老鼠大概2万多一个strain. 国内几个代理都是这个价
钱。时间3个月左右。
还是相当贵啊。不过他们提了个很好的提议，就是如果井口很多品系，可以冷冻老鼠精
子，用液氮罐将所有的精子一次运回，费用2万5人民币，然后在他们那里再复苏成动物
，每个strain 4000人民币左右。这样就便宜多了。 不知道采集冻存小鼠精子的过程复
杂不，我想学会，这样就可以省很多钱了。
国内养老鼠也很贵，因为学院没有自己的老鼠房，得租用别的学院的，距离远不说，价
钱比美国还贵，大概一笼老鼠每天7元。我说弄100笼老鼠的话，一年就是将近30万了。
不成就得钻空子自己在实验室里架设老鼠架子自己养。
楼上说的jax在武汉建立中心的估计啥时候能成啊，成了那真是天大的利好！

a strain of bacteria may emerge that no antibiotics can kill it.

more than a dozen classes of antibiotics, including hundreds of drugs in
a mere 70 years. But even such a day does come, it simply brings us
back to the pre-antibiotics |

(2) You need to consider many parameters: modulus, strain and stress.
check your Experiments for Polymer Science edited by Ou Guorong.
There is one experiment for that, very detailed.
stiffness is judged by modulus;
toughness is judged by strain;
strength is judged by stress.

thickness. therefore for the the entire laminate, there is no separate
DOF for each layer. only that the integration is done for each layer to get
the appropraite K. after getting the solution, then back calculate the
stress/strain at each integraion point at each layer.
read through theory manual, and you will see how it is treated. the laminate
